The Speedy Goon is the one wearing all blue (knit cap, plaid shirt tied around his waist, sleeveless shirt). He's also in the picture for this achievement if you need a visual reference. Beat on him with any character you like until you get him low on health, then double tap the left stick in any direction to run. Once you're running, press either 
or 
to execute a running attack. There are plenty of these blue Goons in the game, so don't stress out if you don't get it the first time.
Getting this achievement will also fulfill one of the Challenges towards unlocking the Blue Belt achievement:

An alternate strategy if you are struggling with running attacks is to tag team with a different fighter (i.e. Switch characters). It is a lot easier that way.
