Wolves spawn when you spend too much time in an area without moving into the next level. The time required is not fixed. When they start spawning, you will hear wolf howling sounds, and a comment will appear above the player.
To unlock, just spend a lot of time in an area before progressing to the next. Took around 7.5 minutes for me on 1-1.

The more enemies left in the current area the longer it takes for the wolves to start spawning. If you clear a level first, or go back one screen to a previously cleared one, you'll find the wolves quicker.
