The quickest way to get this is to build a house on an empty estate and then wait for it to get infested.
Once it's infested get one of your workers to kill the cockroaches as they come out of the building.

The achievement title is a little misleading. You can 'knock out' or kill any enemy in order to unlock this achievement.
By far the easiest way is to wait for the zombie outbreak after 2 residents die on a single estate. This will be slobs if you set them to produce workers and tenants from the outset.
Find your nearest build crew and use them to target zombies. You will have to micro manage your builders. selecting each one to attack a different zombie. You know when the zombie has been killed as a coin will pop over his head once it is dead. Repeat this for 4 more zombie and the achievement will pop.

Select a worker (not the Foreman) and make him attack a rival worker walking down the street. When you see one walking around, you can attack them with workers by selecting the worker (press 
) and giving him the command (press 
) to attack the enemy rival worker.
Note: This trophy is GLITCHED in a good way in v1.00.
Instead of workers, you can kill 5 Cockroaches or Zombies, in the same manner and the trophy will pop for the unpatched versions of the game so if you have the retail version, you can just play offline without the patch installed but for the digital version, you will have to use the patch downgrade exploit to revert the game to a version prior to the patch which fixed the exploit: LINK
