tl;dr: Max out attack/focus/berserker and EtherSnipe in Wave/Neutral. Max SP. Eat Spicy Beat-O-Stick. Jump in water until HP is red to activate beserker. Find Shokat in Gaia's Garden. Blast with EtherSnipe bouncing off a wall. Pray.
EDIT NOTE 4: StolenPaper18 says that you can start New Game+ and turn on Sergey Hax. (It turns on super damage. Hit something and it should be done.) I will leave the rest of the guide in case anyone wants to do it "normally."
EDIT Note 1: It has to be 50k in one hit. The cumulative counter above an enemy does NOT count.
EDIT Note 2: According to TonySki, keep boosters off.
EDIT Note 3: Consoles currently don't have NG+, so that's not an option (for now).
CONTEXT:
This solution is meant to get you started. There is a lot of different equipment and many combinations might work; I am just showing you what I did. I finished the main game and wanted to NOT farm any additional equipment to be able to get this achievement. I did research to find the best method, but nothing was definitive. I was lucky enough to only need to distribute circuit points, buy one food item, and find the place to do it. Throughout the game, I did NOT craft ANY equipment - I only bought the generic stuff and used what I found. It shouldn't matter, but all my difficulty sliders were on the left(easiest).
ATTEMPT STEPS:
1. Fast travel to Gaia's Garden - Jungle Entrance.
2. [Save here.]
3. Exit north to Crossroad, defeating enemies to build max SP.
4. Exit east to Splitting Stumps.
5. Beat the Shokat, continue east, beat the plant thing, and eat the Spicy Beat-O-Type.
6. Jump in the water until you have red health to activate beserker.
7. Find the group of Shokats and fully charge the Ether Snipe and try to bounce it off a wall (or 2 or 3) into the Shokat.
8. Pray.
9. Exit to menu and reload if it didn't work.
RESULTS:
The first time I hit the cat, it was like 48k. Next try was 23k. Then 94k. So it seems very variable, and you might need a crit or RNGsus.

So we don't have a NG+. The epilogue only lets you go back to the beginning of chapter 10. We don't have the Damage Boosts from Sergey outside of those 2 boss fights. Even then you're hitting insane numbers but the achievement isn't unlocked then.
I'm having issues with it because I'm level 65 now, the cats were too annoying to try to hit so I went after the Hedgehogs in Autums's Rise. First bounce shot with berzerk mode I got a 200K hit counter. But No achievement. Multiple times I've gone over 50, 100, and 200K with the hedgehogs, the Pandas in Sapphire Ridge and the Bug Samurai there as well. No achievement and the in game tracker for One Punch shows my highest so far is only 43K. I do have some late game equipment so I should get it easier than you but it's just not going.
EDIT: FINALLY GOT IT!
So Berzerker with the expensive food and used the beetles instead. The issue was that I was fighting with the Booster on. That stops the achievement from unlocking for some reason. Just used the Japanese Bridge in the center of River Road to get a triple bounce. Had to be quick as the bugs like to be on the same side of the bridge as you though.
TLDR: LEAVE BOOSTER OFF

Start New Game+ and turn on Sergey Hax. Easiest way to get this.

The, Pants, Party
This is the only achievement in the game that is not story related. You are required to do 50k damage to a single enemy in a single hit (in other words, an attack that hits multiple enemies at once and adds up to 50k total won't work). There are a number of ways to go about this, but the best method I found is outlined HERE. Essentially, you will be equipping gear that relies on the "Berserker" skill, which raises your attack greatly when you are under 33% health. Then you'll want to spec your Circuit to maximize the damage and critical hit nodes. If you're doing this after the story is complete, you should be a high level with plenty of stats.
Once you think you're strong enough, get into battle (dismiss any party members first so they don't kill the enemies) and jump in water to lower your health into the red. You can also eat a "Spicy Beat-O-Type" food item for a big boost to your stats right before attacking. Then unleash your most powerful attack on a weak enemy in Autumn Hills. There are skills that will do more damage such as Ether Snipe or Vagabound (especially if you use one that has bonus damage after ricocheting), but any Tier 2 or Tier 3 skill should do the trick.
Note: There are a few story spots where you will get a power upgrade and be hitting for hundreds of thousands of damage per hit, these do not count toward this achievement.
