This will come with natural progression if you are going for all of the other achievements.
You could probably focus on finding the Blacksmith and spending soul gems to increase your chance at better starting weapons. This might help you kill enemies faster early on.

This is likely your last trophy. I needed about 300 kills (check via touchpad /trophies)... endless mode does not count for kills (discovered this the hard way). The way to do this is to pick the third mission (water level)..your goal is to get to the final stage, where there are 3 pillars you're supposed to touch. Enemies will spawn endlessly, and I'm fairly confident this is significantly faster than any other way.
Create a build for quick kills of mobs. I recommend an axe. There are many ways to do this, though I got lucky with invisibility (lets me escape and reposition), a relic that gave me +10mp per kill (possibly the most important), and free electricity hits per break (or kill?) That let me cause chain reactions. I used the Demon Lord (I think... It's the one who lets you have fire hit all around you for 10 turns and jump stun). Given I could recharge per kill, I was invincible.
Still took a bit to grind 300 kills... But far better than any other method I can think of. There are plenty of other relic combos that might work but I do think the axe is essential for group hits. And even if you have a bad luck run and die in the room before your done, even 50 kills in this room is fast progress.
