Once you get to Wishuel after the introduction, you'll find the Cathedral, which will allow you to save, view the gallery of images from scenes you've seen or buy prayers. Prayers are temporary stat increases that affect your party for one dungeon (such as increased EXP or DEF). If you want to unlock this trophy without spending any money, you can purchase the "Only the Goddesses Know" prayer, which is free but puts a random effect on your party. You can only buy one prayer at a time but if you want, you can easily grind this out by traveling to a random dungeon and returning to town for another prayer. Just be mindful to get a prayer (even if it's the free one) before going out of town on a quest and this should unlock after a while.

In the Cathedral, you can select to use a Prayer. Most Prayers cost Bells to use, but there is one that costs nothing, but the effect is random. The Prayer's effect wears off once you leave a dungeon. As the trophy says, you need to use 50 Prayers. You can use the one that costs nothing, so use that one every time you go out questing, and you'll earn this easily.
