When you first meet the priest in the temple of Pentatonica, you can challenge him to a duel. Scroll down a little to find that dialogue option.

If you missed it the first time you talk to the priest, you need to start a new game for this.

I skipped it the first time by accident and exhausted the dialog. The option to duel him wasn't there anymore, but I loaded up my autosave which put me outside the temple again. Walked in and could choose the dialog option again. Didn't have to start a new game.

When you head to the Pentatonic Temple for the first time, relatively early on in the game, you will head inside and talk to the Pentatonic Priest. Before proceeding with the story at all, you must scroll down to the bottom of the dialogue options and challenge him to a duel. This option is only available the first time you come to the Temple to talk to the Pentatonic Priest, so if you forget to choose it, you'll need to start a new playthrough.
