A Finishing Move is when you have an enemy subdued but still active. You can walk up to them and press 'B' and it will do a little animation of you finishing them off.

The standard enemies (Men with guns/hazmat suits) can easily be prepared and killed with a finisher. Just throw your glaive at them (Not a power throw) and 9/10 they will stagger and have a read outline. Run up to them at any angle and you'll be prompted for a finisher.

To perform a finisher, either get behind an enemy or hurt them so they are glowing red, get close and press 
These should be fairly easy to come by throughout the game, especially on zombies as they have no ranged attacks to deter you.
Use the section described in "Headhunter" to rack these up quickly. You can also get a bunch during the first sighting of the Colossus because the enemies will be shooting at him and you're free to walk right up behind them and kill them.
