this as many other achievements can be obtained cumulatively in several playthroughs, as long as they are NG+ on the same save slot

This will come naturally over time. Be forewarned that the second I killed the enemy that gave me this achievement, my health dropped to 0 and I was killed. I'm not sure if this is a glitch, or an intentional joke by the developers.

If for some reason you don't unlock this naturally through your play through - go farming in the Cathedral graveyard.
Redemption, Mode
This will come naturally as you progress through the game. Even if you die and cleared an area of enemies, the kills still count as you can clear the area again.
