Trying to get this on multiplayer against humans is damn hard, but by comparison, playing single player against bots is a walk in the park.
Personally, I recommend (for all boosting), Pier 70, free for all. Its a small square map, and that way I was finding I could get between 50-75 kills per session.
Just run round like a lunatic, pushing down RT to knife (only AI enemies, NOT zombies as they don't count). The AI is so poor at shooting, you can just run at them, and still get them before they shoot you. Play as light class, as you can run faster.
In between deaths, don't forget to hit LB to throw a distractor (x3) to boost this, and Y twice per life to also use your other equipment (varies per class) to rack that up as well.
