The map screenshot is no longer terribly helpful, but this should show you what you're looking for if you've never seen one. Note the faint glow at my character's feet, as well as the text letting me know I can use the rune.

Sadly, I think the seeds change on every update, so I don't think your seed is good for this anymore.

It looks like you're right. I tried that seed after I read your comment and the map has changed. There is no rift in Prisoner's Quarters, and I didn't see one in the Promenade either. I will make a new note in the solution.

For this one you need to find a Challenge Rift, open it, and finish it. It has a 20% chance to spawn in a non-boss area, and will have a prompt when you run over it to interact.
