First you have to do a run where you go to Castle's Outskirts -> Dracula's Castle -> Defiled Necropolis and defeat Death there.
Then, to access the Master's Keep, you have to take the regular path to the Clock Tower (don't go to the Castle's Outskirts) and you'll have a new door after beating the boss in the Clock Room to go in an alternate version of Dracula's Castle which will let you access the Master's Keep.

It’s worth noting that you need to go to the Castle’s Outskirts and then to Dracula’s Castle/Defiled Necropolis to beat Death before any of this is possible.
