Unlike the 4 previous boss cells, it's actually the Giant who gives you the 5th, NOT the Hand of the King. With this in mind, start the game with 4 boss cells active and make your way to the giant (if I remember correctly, I think you need to go through Toxic Sewers, Ancient Sewers, Insufferable Crypt, Graveyard, Cavern then Guardian's Haven. The probably are alternate routes too.) After beating the Giant and getting the 5th cell, you can go to High Peak Castle or skip it completely and head straight for the Hand. Beat him, start a new run and activate that 5th boss cell.
Now make it back to the Hand and beat him again. With 5BCs you can finally open that locked door past the fountain, which will take you to the Astrolab.

Make sure you pick up the cavern key in the beginning of the 4BC run, made that mistake myself

You can unlock the cavern at any time (when you have all the base runes anyhow) nd once unlocked it will stay unlocked.
