The Derelict Distillery is accessed after Clock Room and beating the Time Keeper, as long as you’ve got to the Throne Room once before.
Most ‘normal’ runs will take you through the Clock Room, just avoid the Caverns route.
For example:
Prisoners’ Quarters->Promenade of the Condemned->Ramparts->Black Bridge->Stilt Village->Clock Tower->Clock Room->Derelict Distillery
Obviously you'll need to make sure the Derelict Disterlly update is installed.
