Gas Station south. There's a Tunnel where infected will continuously Spawn. since Infected dies in 1 hit you can wait for them to show up one by one and kill them for the achievement.

(If they somehow stop spawning, run twords the gas station and back to re-spawn them)

Note: Remember only kill Infected, hitting any other zombie will reset your kills for "One is all I need"

If you can create Developer's No. 4 Craft, you can definitely unlock it.

bake a purple skull on the stove. get Developer's No. 4 Craft blueprint

I found this achievement easiest to unlock during the main quest Only the Strong Survive at the end of Act I. This is the quest where you'll meet the recently bitten mechanic Earl and his daughter Jin. Earl know's he doesn't have much time until he turns but is still well enough to help you reinforce the truck you arrive in. With all the welding needed, this won't be a quiet job so you are told to protect the workshop while he works. Not long after you go outside, the Infected type zombies (the fast, viscous you can hear from a distance) will start running at you from a distance in either pairs or fours. In total around 18 zombies will attack you so this is a great spot for this achievement to be unlocked. I found that using analog controls were almost easier, but I was able to do it with the digital as well on a different profile. A Bleeding Baseball Bat and Machete both fully upgraded were the weapons I found easiest to use. Although a little harder to find, a Katana will also work just as well. They will be coming at you a little fast so just be prepared. If you fail to get 5 in a row you can always reload the checkpoint however, med kits, weapon conditions, and health do not restart so just be aware.
