How to unlock the Perfect Gunner achievement in Dead Rising - Definitive Guide

Later on in the game you will get a machine gun quite easily (not sub Machine gun). When you do go into the meat locker through the garage below Seons in the north west corner of the map. Shoot a hanging meat without missing and this achievement will unlock! Make sure it's a full ammo Machine gun.

Just to add on to Strategy Ones' Solution,

The machine Gun Has 150 Shot when it hasn't been fired yet.

you can also skip 72 hour mode if you have already beat the game and start in overtime and still get this.

there is no recoil when shooting so jump on one of the tables so the zombies can't get you and fire into the hanging meat in the butchers ( thru the maintenance tunnel ).

You can pick up a machine gun from any special force guy later in the game. Just make sure it has full ammo before going to the butcher.

This must be done with the Special Ops machine gun as far as I know. Grab a few and head into a highly populated zombie area. Corner yourself and take aim. All 150 bullets must hit, so take it one at a time and take your time. If you miss, switch to another fully loaded gun and start over.

You can also achieve this towards the end of Overtime mode when you are escaping underground. Towards the end there is a jeep that you must escape in with two Spec Op guards nearby. Kill both of the guards and then take the machine gun off the jeep. With the machine gun from the jeep, shoot the jeep until every bullet is used and you should receive the Perfect Gunner achievement.

The machine gun, the one that falls out of the special forces soldier, take it and run to the Meat Shop , which is located in the Service Tunnel . We aim for a piece of meat hanging on a hook and shoot all 150 rounds at it, after which we get a trophy.
