How to unlock the Now With Sprinkles achievement in Defense Grid 2 - Definitive Guide

Tower augmentations are buffs you can earn for each of your towers, which you select at the beginning of the level on the screen where you choose your command team.
As far as I can tell they are randomly earned, based off the fact you can trade them on Steam. So your best bet is to just play through the game until you get one. If you earn one when finishing a level, they show up on the left side of the screen. They'll also be marked by a ! when you're choosing your command team before a level.
To enable one, simply scroll down to a tower you have earned one on, scroll to the right to go to the list, the choose an augmentation.

As with all other achievements in this game, this will not unlock until you complete the level.

As you earn medals in the game you will be rewarded with tower augmentations that enhance the combat abilities of the various towers. When starting a game there are three screens to go through: Mission Select where you choose which map to play on, Mission Options where you choose the game mode, type, and difficulty, and and the Loadout screen where you can assign tower augmentation items to the various tower types.

To unlock this achievement you will need to choose any one tower augmentation item on any tower while on the Loadout screen.
