How to unlock the Witch Hunt achievement in Destiny - Definitive Guide

In the Crucible, simply kill enemy players. Once you kill 100 Warlocks, this achievement will pop. The type of death is irrelevant, can be by weapon, melee, grenade, super, etc.

As long as you get the "Kill" points to appear, you get the credit.

Note that Assists were confirmed to count. So either the "Kill" or "Assist" message needs to pop, both will count.

As of the Rise of Iron DLC update on 9/16/2016 you can now obtain this in private matches.

Play PVP Matches which are called the Crucible in Destiny.
Every Time you kill enemy Players (Kill +100) or even assist in a Kill by your Teammate (Assist +50) it will count towards your Total.
In the Heat of the Action (for me at least) it's hard to tell which of the three Enemy Types (Hunter, Warlock, Titan) you've killed.
So maybe it'll help you to go to the Bounty Board and get some of the "kill 25 hunters" Bounties, which will be available again, a short time after you turned them in.
So you won't need a total if 300 kills to get yourself those 3 trophies, cos assists count as kill as well.
How fast you'll get those trophies depends on your level of skill and luck (and your weaponry) but even for a mediocre PvP player as me, it doesn't take long.

I believe this needs to be done with one character. I have over the 100 kill count on my grimoire card for each class, but these counts are across all characters, and nothing has unlocked yet.

Playing Control is your best bet for maximum kills. For some reason, even assists count toward a successful kill for the achievements, so shoot at everything you see even if it might mean your death. A teammate avenging you will still count. While going for these kills, gather the heavy weapon ammo and use your special ability to try and get "Excessive Force" and "Triple Play" out of the way. Also try to work on any Crucible bounties you might have, and earning marks to max your weekly count.

This is a Crucible grind. You can begin this after the Crucible unlocks at Level 5. As long as you put in the time in the Crucible, this will occur naturally.

This can be done in private matches, you will just need to keep killing Warlocks until the trophy unlocks.
