How to unlock the Headhunter achievement in Dishonored Definitive Edition - Definitive Guide
The 2 Most Diffucult Rooms are 4 and 5
• 4th Room is tricky, I personally let the assassin swipe his sword at me when hes really close up Then go for it
• 5th Room is all about Patience, Accuracy, and Timing... for the assassin going up the stairs.
• Really you want to find the EXACT SPOT to aim in this video.
Here's a Visual for this achievement, although he got lucky in room 4... feel free to use the up close method.

Tip for the fourth room. Before you enter the room you want to use Dark Vision to tell you which door has assassin's behind it. You then want to Double Zoom and aim for the head of one of the assassin's and slowly inch into the room. If you only just step into the room, you will have a brief, very very brief second in which to head shot that assassin you were aiming for.
Then run into the room they come from and climb onto the back drawer/bookcase and sit in the corner. The other assassin will try hit you with his darts or pull you out with his vaccum but they won't deal much damage. Eventually he should back himself into the corner, so let him take another shot and then quickly regain your aim and pop him in the head. This worked for me 9 out of 10 times and for me it was a lot more reliable.

The only tip I can offer other than what's already been stated is: There is a luck factor that plays out here. Just aiming at the head sometimes isn't good enough. Your reticle dot will turn red when aiming correctly but lots of times the hits don't register, forcing you to restart. My advise? Try, try, try again and again. Brute force will eventually carry you to the achievement.
