Therefore, I recommend expand room size to hard or nightmare and stage 1-2 rinse and repeat.

I'd also like to add that you can set it's to random not mastered. Each run you'll use something that needs to be mastered. No need to keep track. I'm just aiming to max as much as I can before I really dig too deep with a special set.

Adjusting the loadouts is very helpful for this, as stated above.
One extra tip for quick clears: try to find the enchantment room ASAP and dump all your sand into your melee weapon. Using the heavy attack (Y) you'll be able to one shot all first floor enemies.
