You will want to get this out of the way in the first couple of waves.
Just purchase the lvl1 sword and get close to the enemies. In the early waves its usually a 1-2 hit kill with the sword.
Kill 5 with it and then swap out for a machine gun.

This should unlock during Wave 3 if you have the sword equipped from the start.
Before starting Wave 1, access the store by pressing
. Select ‘Glaive I’ for both the left and right modules, then press 
to start the wave. There is one enemy in Wave 1, two enemies in Wave 2 and three in Wave 3. To destroy them, just keep pressing and/or while next to them.
