Mana is the game's secondary currency and is used to buy certain upgrades like increase your dungeon population, research spells and cast them. To gain mana you need to first have a mana crystal to mine off and this aspect is unmissable as part of the campaign. The game also teaches you how to mine mana so it's fairly simple. In any case, crystals can be found in most story, skirmish or online maps, and can be seen as blue veins in rock, or as large floating blue crystal formations. To collect mana, build a Mana Room on and around the Crystal, then send either a Naga (Orc faction) or a Shadow Lurker (Demon faction) to the room and they'll begin channeling and collecting mana.

It's unlikely you can get 250k mana from simply playing the campaign and will need to replay campaign maps or try a few Skirmish or Online maps to finish this off. I recommend The Poshlands Skirmish map as it has 4 mana crystals to mine. Keep mining until the trophy pops. You will need to make really large mana chambers to hold everything so either spend mana by casting spells or keep enlarging your rooms or simply complete the Skirmish and restart, since mana mined is cumulative across all sessions. There's a spell called Gold to Mana that converts mined gold into mana but it's unclear if this adds to the cumulative total for the trophy, since there's no way to check how much mana has been mined thus far.

I also recommend playing as Demons if playing Skirmish, as the Demon minions have the teleport upgrade allowing minions to teleport quickly from point to point, making general work a lot faster. You also might want to upgrade your Shadow Lurkers (used by Demons to mine mana) via the Summoning Chamber Pentagrams as upgraded versions mine mana faster.
