This gun is found rarely and with little ammunition. A few Heavy Armours will carry these. It won't always be a one-shot kill so you should shoot and injure replicas (or nightcrawlers) with another gun first. This must be done in the Perseus Mandate campaign. It has been confirmed that this must be done in one playthrough (thanks to D4RK I FuRy I).
A good way to horde more ammo for this gun (AND EVERY OTHER GUN) if at least two enemies dropped it is to take the gun, then SWAP IT BACK before you take the second one. This will give you MORE ammo than if you were to pick up the second one immediately after the first one.
This is because the game gives you a bunch of ammo if you pick up a different gun, but less ammo if you already have the weapon in your inventory.
Example:
if you swap your pistol for the Penetrator, you'll get 75 rounds. If you pick up a second one you'll get only 10 more, giving you a total of 85.
But if you had swapped back for the pistol and THEN picked up the second Penetrator, you would have had 2x 75 rounds, totalling 150 rounds!
NOTE: these must be weapons DROPPED by enemies, not the ones that are lying around for you!

Does it add up over multiple playthroughs? I didn't get it in my first playthrough. Or do I have to kill 20 enemies with this weapon again?

Not sure, but my guess is that it won't stack over multiple playthroughs. Try to make every shot a killshot in your next playthrough just in case. I fired one torso shot(burst) with the ASP before using the lightning gun at point blank range. If I'm wrong you'll get the achievement sooner.
