You need to have 50K population in a single Colony. In unlocks several days after.
No need for the colony to be able to sustain this population, just ship them back once the achievement is unlocked. (takes a few weeks)
Main difficulty is getting enough population in the main city to be able to ship out over 40K people and not cripple the city.

I did this on my 60K population playthrough and just sent 50K people to one of my colonies, they had no housing, and it crippled my main city, but hey, it's a harsh world out there. Some people died, and it took it down to 49K (at that colony, I had other colonies), I went back to the transfer people screen and the achievement popped immediately - so that's worth trying.

Play the utopia to make a huge city reach the 50K to end mission and then continue to play then just send all of your main city to another city.

I'm also got a notification about my character dieing in game in 100 or 150 weeks so there maybe a hard end on the utopia.

It appears to only count colonies, not outposts. I tried in this in the main story on the embrace path, and all outposts fully upgraded should be 18,200. But it only unlocked for me when the 3 colonies had 50,000 between them.
