I suggest doing all the level specific achievements on your easy run and/or with modifiers for a faster completion. At the end there are 3 waves of enemies with friendly turrets. Wave 2 and 3 have one of each of the boss ladies. DO NOT SHOOT THEM. They will leave once you kill all the B.O.T.S. Just focus on ads and avoid killing the bosses.

As destro you’ll make your way to the big platform you have seen in way to many missions. You’ll see several turrets you can activate for the upcoming 3 wave defense section. First wave is just various robots. Wave 2 lady jay appears with the various enemies. You can see where she is by her asterisk symbol on the mini map or the gi joe symbol. Just run away from her and kill regular scrubs from far away and once last one dies lady jay vanishes. Rinse and repeat for wave 3. Scarlet appears but will charge you at start. Just stay away from her and kill all the regular enemies. Once they die scarlet runs away as well. I’m not sure if the turret kill on her counts for this so I would just avoid using them.

Make sure you're playing as Destro. During Mission 13, Lady Jaye will show up during a wave of enemies and then Scarlett will show up. Ignore them. Just run around killing everyone else and they'll eventually go away. Mostly you just want to avoid getting shot too much by them. They aren't the most powerful of characters and deal little damage, but if you're struggling, use those modifiers.
