Now with the latest update by TC removing the Custom Escape maps , they have very nicely gave us another unwelcome grind for the 100k kills on Horde/Escape.
Currently i am using the map "The Detour" running Brawler class. Upon spawning i make my way to the end of the hive in the first chapter, bypassing a few of the safe rooms. Along the way you will either come across (Drones, Buzzkill Scions, Rejects and Leeches), or (Drones, Ice Scions, Rejects and Juvies) which are simple enough to deal with.
As you come to the very last Saferoom turn left and you come to a deadend, from which either Leeches or Juvies keep spawning around 7-8 at a time. I would recommend using the Brawler (My loadout is Defensive Tackle, Perk up, Soothing Warmth, Torch tackle and All the glory).. as you can use your tackle to kill them all, then rinse and repeat for a few minutes until the Venom catches up to you..at this point the enemies stop spawning.
I then just restarted act and done same thing again...on average i got around 700-800 kills per run with Leech spawns and 300-350 with Juvies, so i'd recommend Leeches.

I suggest doing all other Horde/Escape related achievements first as you'll be working on this one at the same time. if you still need to do some killing or just want to do straight kills then this is the best method I've found.

I've been Averaging 1350 kills every 12 mins. FYI I've added in a safe room at the start so that you can restart act and not have to sit through the cutscene each time. kill the 1350 enemies and then restart act.

Fastest way to do this is to load up a custom escape map (Escape > Map sharing > 100K Farm > Host with this map) with alot of enemies and alot of heavy weapons for you to kill them with.

I made the custom map "100K Farm" ID: SCHF51. It's 12 bays of 200 Juvies each. each room has 4 salvos in it. I suggest playing as Clayton as he heals everytime he does damage with a heavy, but you can use whatever character you want, and the juvies are set to aggressive for speed. Enjoy.

Note: You don't get XP/CXP/Cards in custom. this is straight for cheevo farming.

I wanted to share my setup that helps me get around 900 kills every 10 minute. You need to pick ANCHOR class, start a private Escape match on DETOUR and get the LEECH spawn.

After breaking the pod, go in to the first supply room and get the ammo. First batch of drones will be entering the supply room, clear them, then there will be some leeches coming at you. After dealing with them you will get 3 rejects and 2 drones coming at you. Clear them too and you should have your ultimate available. Use it when you see any other leeches or enemies. If you have the below setup you will run through enemies easily since barrier will kill almost all of them instantly. And every dead enemy will extend barrier time at least 2 seconds. THAT MEANS ALMOST INFINITE BARRIER TIME. Now go all the way to the opposite of the safe room where the leeches spawn, clearing any enemy that hit your barrier along the way. At this point it's entirely up to you where you can make your character stay put but I personally stand at the side of the entry location to the spawn point. Now you can grab a coffee or tea while leeches drop dead while hitting the barrier. Occasionally some will surpass the barrier and explode on you but it shouldn't kill you. Personally I found this setup a little bit better than Brawler method because you can basically go AFK for about 7-8 minutes with this method.
