After you defeat each boss, you'll unlock two abilities: 1 active and 1 passive. After you complete the game the first time, you'll unlock the rest. You can (and should) do this trophy in Arcade Mode. In Arcade, you can pick and choose which bosses you fight and will have all of the abilities available for use. Just keep cycling them out after each fight (you don't have to win the fights, you just need to start them) and once you've used all of the abilities at least once, the trophy will unlock. 

For the active abilities, you need to actually physically use them. This will of course require you to fight back and get some souls but it shouldn't take to long to get through them all.
