At the start of Chapter Two, Alfred will mention about a raid on the S.T.A.R.S labs once you're in the city.
Head there (marker is in the South-west) and complete the fotress-style level and this will pop once the game tells you to go back to the Belfry. Go back there to continue the storyline.

After completing Case File 01, a new Case File labelled “?.? - Heist at S.T.A.R. Labs” will become available on your map.
Travel to the lab, fight off the Regulators hired by Mr. Freeze, and complete the level to unlock the achievement.
