This mission is from Salvadore (The "S" on the map) after completing "Bomb the Base II".
You will be told to get a car from Luigi's place to take to the crusher, but will get a pager message once you get close to his club. Do not get in the car, or it will explode and you will fail the mission. Drive to the Pink square on the map to meet Maria. She will introduce you to a new partner, Asuka. You'll then get on a boat, which you will take to Staunton Island, unlocking the second island.
The next time you get into a car, the radio will tell you the bridge between the 1st and second islands has been repaired, allowing you to travel between them.
