There is no need to read any other solutions or look anywhere else on the internet. This achievement is actually one of the easiest ones, as long as you know what to do. For the first two rounds just lock onto your targets using LT and just click B when they are close to you. If your health isn't full after the 2nd round, choose to spectate after the round to revive your health(*this applys for every round). Your next round will start after you watch a match. In this 3rd round the second guy will come at you with a bat and the third a knife! But dont be scared all you need to do is counter. Now you have a weapon and can easily win that round. Your Weapon dissapears after each round(as does it if you defeat a opponent without taking his weapon, you cannot pick it up off the floor).
If you remember these few simple tips, you should be able to unlock this achievement first try:

Here is a useful trick I used that made getting this achievement extremely easy. Lock onto your opponent using LT and just keep bashing X (kick). This works because Luis can kick faster than the opponent can recover from the attacks, meaning that you can get them locked into an endless cycle of kicks. Plus there is a little lunge kick Luis performs so you can catch them off guard when you are just out of range. If you are doing it right, they won't even get a chance to fight back! This also works in the later rounds when the enemies carry weapons too, if you're not good with your timing and can't dodge attacks very well, just keep kicking them and they can't hit you. You don't need a weapon if they are constantly getting bombarded with kicks.
The only downside is that the kicks are weak and do little damage. However if you persist and have patience then the achievement should come easily. Another tip is to lean toward the enemy with your Left Stick slightly, don't move it all the way, this helps you to stay within kicking range and that you don't miss a kick. After finding out this little trick I got the achievement on my first try, good luck!

I combined the advice of both LawL PwNeD and DeafAtheist and won the Achievement on the first try without breaking much of a sweat.
DeafAtheist - going for first blood and catching opponents coming in with tap of the X button, is good advice for taking them off balance (especially the ones who come charging in at full speed) as well as scoring some fast damage, which against the tougher opponents in the later rounds, setting them up to be taken out easily with the GTA fighting combo.
LawL PwNeD - Follow that up with the GTA fighting combo (tap A button and pull back exact at the moment an opponent attacks; followed immediately by tapping B button three times and pushing forward), usually took care of almost every opponent. This combo will also disarm any opponent with a weapon, at which point you can use the weapon against them.
LawL PwNeD - If things don't go 100% right and you wind up taking more than 50% damage, you can use LawL PwNeD's SPECTATE trick to refill your health meter to 100%, before taking on the next round.
- SOME NOTES -
1. If you have an older, worn down controller, no matter who's advice you take on this Achievement, it is still going to be pretty hard to win. Using the combined method above, I tried winning the Achievement again for a friend on his Xbox 360 and his controller. His controller was more worn than mine, and after half a dozen failed attempts, I switched from his (older) controller to my (newer) controller, and nailed the Achievement on the very first attempt.
2. The spectating mode is mostly rigged. There is a pattern to who wins in each and every round. Pay very close attention and learn the pattern if you want to earn big bucks. Then without resetting the system or the game, reload from a save, and bet accordingly. Resetting the game or the console, will reset to a different pattern. On one play-thru, I started with only $25,000 and ended with almost $700,000, simply by learning the win-lose pattern of the Liberty City Cage Fights.

The achievement can be gotten without spectating between rounds as LawL PwNeD described, but it's challenging. It took me about 20 or so tries to do it.

I found the best way of doing this was to kick each fighter in the jewels as they approached and then hit them repeatedly until the went down. Few of them barely even got a punch in before being dispatched.
Once fighters start coming out with bats and knives it's harder, but the same idea works... kick them in the jewels before they can use their weapon against you then take a couple steps back, duck when they strike then disarm them and use their weapon against them and the rest of the fighters until the end of the round.
If you start losing a lot of health sometimes it's better not to try to duck and disarm because sometimes you don't duck in time and they can take you out very quickly. If your health is getting low just get on the offensive and put them down before they can strike without trying to disarm them. When the round ends you can regain a bit more health to keep fighting.
If you have 25% or less health left at the end of round 5 then you might want to step out and spectate before starting round 6 if you have to but it gives you a sense of accomplishment to win the championship without stopping.

A really good strategy and almost a glitch is this. As your opponents try to get through the door, start wailing on them . If your lucky they'll fall into the ring and then for some reason they won't fight back. If they walk into the ring no drama, you've still got a few good hits in on them.

I had this game for awhile and i just start to play it again, this how it works for me on my third attempt. the opponents are basically the same even the ones with the weapons, the only difference is that they can take more hits as you progress thru the rounds. just by keeping them at distance hit them with the x once the long kick or the knee will do the trick and then hit them with b 3 times step back and go again make sure that the kick hits them before attempting to hit them with the b specially the ones carrying weapons, do the same on every single opponents, be patience keep them in distance and you will get it easily.
Keep in mind that there are 5 rounds and the final witch would be the 6th each round have 3 opponents.
....And if you are taking more damage than 50% choose the SPECTATE option they give you at the end of each round that will regain your health to the max, since youll have to do the 6 rounds in one go, to get the achievement.

All you have to do (If you want to win without taking any hits) is just kick the shit out of them, constantly kick and they wont be able to hit back. I just worked this out and it's such a stupid thing but it works.
Rounds 1-3 you can just punch them with B constantly, but rounds 4-6 you want to stand in the far corner opposite the gate, let them run to you and then kick constantly (if you kick them at the gate you will go out of the ring). If you get them in a corner and kick them it's even better because it's a lot faster.
There you go, go and win!

Bundle (leg - shortens the distance) + - works on all types of enemies, especially the most dangerous ones with knives. The main thing is not to forget to move away after each combo, and wait for the moment when the enemy runs up to a foot's distance.
