You will have 1 Assassination Mission available at a time, after completing them the follow-up ones will become available. Each time before they become available you will get a phone call telling you where you can find the phone booth. To get the final mission available you will have to meet more requirements.
Loose Ends, how to unlock:
1. Buy the Malibu Club property, which will cost $120000
2. Complete the No Escape? mission, which can be found inside the Malibu Club
3. Complete the The Shootist mission, which can be found inside the Malibu Club
4. Have Assassination Missions 1-4 completed prior to this
5. After having these requirements done, it will trigger the phone call to get this mission available

If the mission doesn't start when walking into the mission marker at the payphone, try crouching within the marker.

changing weapons from fist to pistol and back also triggered the mission within the circle for me.
