This is not the fastest way, but the easiest way.
Play through the career on beginner/vocals. Just put your mic in front of a fan or another noisy source. This should bag you 6 stars on every song except 2 wich dont have vocals.
Now do the challenges which unlocks cheats, e.g Auto-kick for drums, No-fail, Always slide and all notes Ho/pos. Cheats does not disable achievements! (too bad I didn't figure it out untill I only had 9 stars to go lol)
Note streak challenges can mostly be done on medium or hard, so can whammy and starpower.
Save challenges like hitting X amount of ho/pos and taps for after you've unlocked cheats. Dont bother with the band challenges as you should be able to get more than enough stars this way. The "Challenge of the Supergroup" achievement can then be earned seperatly.
Usefull cheats:
-Always slide(all notes are tap-notes) : Get Platinum on Du Hast
-All notes ho/pos : Get Diamond on Hurts So Good
-Auto-kick: Get Gold on Mirror People

I found out something really major when me and my friend were doing some of these online. If you are playing in a band and everyone is going for say "hit x amount of notes on guitar" challenge. If one person gets diamond, everyone gets diamond, which goes for their respective bands! Which means if you know someone online who is really good at beating some of these challenges, they can get them for you too.

The other guides are very good, but I also found a challenge guide on xboxachievements.com that gives a strategy for each individual gig. These are very helpful for the specific challenges, and they provide a quick reference if you're looking for a specific type of challenge.

As stated above, each song can give up to 9 stars, so you only need 808 of 909. That means if you can get 6 stars on a good number of songs, you won't need to Diamond as many challenges.
And, of course, the mic/fan trick works for all of the songs but 2, so if you're okay with fudging all the songs and wasting your day pressing the "A" button over and over again, you can get 99*6 = 594 stars without working.
You can also get Diamonds on "Steady, As She Goes", "Blue Orchid", and "Incinerate" just by turning on auto-kick/no-fail and letting it do the work for you. There's another 9 stars. Cheats definitely do not disable achievements... I got the feeling that by earning those rewards, the game is helping you along. (I just don't think they intended to help you THAT far along.)
So doing all of that, you should be able to get a total of 603 stars without playing a note. It ruins the fun of the game, I think, but there you go. Only 205 stars left to go. :-)

If you find yourself falling short of the 808 goal because you're not quite good enough to get diamond on all the vocals or drums or guitar or bass or band challenges, keep in mind that FC'ing a song will get you an extra star (6 stars).
I fell short by about 30 stars and resorted to playing 30 different songs perfectly in order to get the one extra star for each of them. Doesn't matter what difficulty you're on either, just don't make any mistakes. It takes longer but it isn't cheating.

If you're having trouble with the tap sections challenge on Du Hast, it becomes much easier on hard with the All Tap Gems cheat. You can earn this by getting Platinum on this challenge, which isn't too tough to do. This cheat will also help you out on other challenges like 21st Century Schizoid Man. The auto-kick challenge also makes it extremely easy to get a bunch of the drum challenges. I recommend using the fan/beginner vocals on all songs first, then eep chipping away at the diamond challenges and this achievement will come in no time :)

Now, let me start this off by telling you that there are 909 stars available. 5 for your score performance (on any difficulty), 1 for FC'ing the song, and 3 for the challenge assigned to that specific song. Since there are 101 songs (85 mandatory songs, and 16 selectible songs), the 5 stars for performance are obviously going to be the meat of your star collection.
Now you can easily FC every song on beginner using the mic/fan trick (Place a mic in front of a fan or equally noisy object to FC songs on beginner) except for 2, which would net you 594 stars (606 if you FC those two songs on another instrument). You're already very close, and you've barely done any work.
After all of this, you should have gotten the 5 Star related achievements, and the "Starstruck", "The Traditional", "Iron Lungs", "The Grand Tour", "Did you Finish Like We Did?", "You Want More", "No Mistakes Allowed", "Chanteuse", and the "Open Minded" achievements, with some other misc. ones that you might have picked up along the way. Congrats, you little achievement whore you. ;P
But that's not what we're after. Now we're after the challenge, right? The juicy 100 GS. Okay, but we're going to have to do some challenges.
Now challenges cannot be completed on Beginner. Normally, the higher the difficulty the easier to 3 star. But it's not ALWAYS the case. Again, the challenges can be done on ANY difficulty besides Beginner. Whammy and starpower challenges are very simple on the easier difficulties, and most note streak challenges can be preformed on easier difficulties as well.

You need 202 stars to tip you over the edge of achievement godliness. Here's a breakdown of what each instrument's challenges will net you if you 3 starred ALL of them:
Guitar: 34 challenges = 102 stars
Bass: 18 challenges = 54 stars
Vocals: 15 challenges = 45 stars
Drums: 19 challenges = 57 stars
Band: 15 challenges = 45 stars
Going by this, you're going to have to have more than just a guitar, or online access for help (as challenges your partners earn also cross over to you).
Also, the cheats you unlock for meeting certain challenge requirements do NOT disable achievements or challenge progress. You can activate them in the character selection screen under the Options tab. Here is a list of cheats that will net you challenges EASIER!

Now for the "Collect an X amount of Star Power" challenges on certain instruments, that has an easy solution. When bandmates have full star power and they hit another star power phrase, the energy floods over to the band-mates. In other words, if you're in a band of 4, tell them to not use star power the entire song except for the instrument that needs the challenge, and all the extra star power flooded over to you will count!!! You could hook up 3 mics and use the fan trick and get every single one of them easily.
Also, for the band challenges, you do NOT have to have a standard band (even though the emblem looks like one, making some people believe that's what you need to be), you can have All-guitars or drums, so that should make the "Get an x amount of points as a band" challenges less challenging as well.

There are a maximum of 909 stars that you can obtain. Say you’re good at guitar, bass, and drums and can play them pretty well on hard/expert. You should not have a problem obtaining all of the challenges for them. If you are bad at vocals and can barely even pass songs on medium (which is me) then if you have a friend that is good at vocals he can help you obtain the stars.
What I am trying to say is that a friend can join your career mode and help you get certain challenges done and if he/she gets a gold, platinum, or diamond then you will get to keep the extra stars that he/she got for you.
Ex. Say you have gold on the tap challenge on Du Hast and you invite a friend to play the career with you and he/she gets platinum or diamond on that challenge while in your career you will get to keep the stars that they have gotten for you.
(Or if you don't have all the instruments, this could also be a help)

What I am trying to say is that a friend can join your career mode and help you get certain challenges done and if he/she gets a gold, platinum, or diamond then you will get to keep the extra stars that he/she got for you.

There are 909 possible stars in career. You can knock off 600+ of them by playing on Beginner and earning a perfect 6 Stars for each gig. You can use the fan trick on vocals for this since it is on Beginner. Then, for the remaining 202, you must play the challenges to earn more stars. Turn on all the cheats you need to assist you for each challenge. Click Here, for a guide to the challenges to make your road to 808 easier.
This sounds really hard to do but with some time and effort you will easily go over 808. You can get up to 9 stars in a songs. 5 for high score 3 bonus and 1 for FC. (Full combo) If you use the fan trick you can get 6 easy stars on every song but 2 songs which have no lyrics. Doing this you would end up with:
83x6, 498 easy stars and you can get 3 more stars for each song. It doesn't up over 808 because of the songs you can choose at the end of it stage. This are probably the easiest to get 9 stars because you can choose a song and it tells you the difficulty and length.
