Simply press LT+A, LT+X, LT+Y, or LT+B
After 500x
The achievement is yours

I kinda did have to grind this in order to unlock it. On my normal playthrough I just spammed the default demo shot any chance I could since it only cost 1 demo point to use. Each mission typically resulted in about 20+ activations per level.

Assuming your grinding for this after finishing the Easy, Normal and Hard playthrough, I would suggest playing ‘No Man’s Gate Revisited’ on G.O.R.E. Great number of enemies, takes 5 minutes, and you get around ~36 Demolitions Shots per run.

60÷5=12 runs per hour, 12x36=432, 432 demolition uses every hour. (depending on if you get more or less then 36 per run.)

If you’ve already been using Demo Shots, it shouldn’t take longer then an hour.
