It appears all of these "in a matchmade game" achievements can be earned in Bot Bootcamp, though you might find this particular achievement easier to do in Quick Play or even Ranked. For this particular one, you need to defend five of your controlled zones in a single game. You will get a "zone secured" and some score if you clear out all enemies who are inside a zone you control. Personally, I had to contest it while doing so (let them get some progress on capping first) and it doesn't matter if you kill them or they leave the zone. What seems to matter is that every enemy is out of the zone, one way or another, while you're still in it.

An easy way to pull this off is with Bot Bootcamp. You have to basically cancel a capture of a control point by an enemy. (The Control point will have a partial ring around it of the opponant's team colour which goes away when you enter it).

In Bot bootcamp, you don't even have to kill the enemies, pick and camp a control point somewhere in one of the levels, let the AI run in and start the cap. If you fire at him enough to pop shields, he'll backpedal out of the control area and retreat until his shields come back. That's your first secure. He'll run back in when his shields go back up, either pop them again or smoke the bot.

you should be able to get your achievement before you hit the halfway mark of a match. The key is to not kill the bots every time they start capping, just chase them out of the control point long enough for you to turn it solid to your team's colour again.

Out of all the maps, i found it easiest to pull this off on Control point C on Recharge. Hide behind the crates or the big cylindrical machine, the bots will tend to run in from the open are entry instead of the stairs, as long as they don't see you they'll run right to the control point.

You can get a zone secured by resetting the enemy capture of a zone your team has captured.
By doing that 5 times you'll get the achievement.

Strongholds game mode can only be done in arena maps (quick play, bot bootcamp and ranked arena playlists) and is selected by chance among other game modes.

Bot bootcamp would be the easiest since bots are easy but if your team is too good you might not be able to get all 5 in a match. Human players do tend to rush the zones more often so if you got a power weapon to clear them out of the zone you might also be able to get it that way.

I prefer a video guide and Silver does a great job showing you how to get this done easily with Bots. It is now much more clear to me what to do so hopefully this helps others.
It is all about stopping other team from taking a zone you control and stepping into the zone after they vacate by any means to "secure" it.

Everyone is saying bot boot camp, but that’s a terrible idea, games are too short and bots are too stupid to understand that they need to capture zones. So you won’t get “zone secured” medals, let alone 5 kills before the game ends. So don’t waste time in bot bootcamp

Play ranked instead. The games are much longer, and players rush the zone hard. I got this within 5 minutes by grabbing a sword and camping zone C. Players walk in and you kill them, repeat until you get 5. You don’t even need a sword. I got 5 more zone secured kills the same game with the BR.

To get this Achievement you must secure 5 zones in a Strongholds game in either Matchmaking of Bot Bootcamp.

To secure a zone let the enemy slightly capture a part of your zone to fill a part of it up with red.

Enter & clear out the zone before it is captured to secure it.

Note that killing enemies in the zone is not securing it, but clearing out the partially captured portion counts towards securing a zone.
