An Assassin is the special Boss Type of Thief enemy. They are fairly rare in the game however the Reaper is still considered an Assassin so he is your best bet for this achievement.
The Reapers Arrow is a Platinum Artifact unlocked the first time you complete the challenge Death and earn the Gold Token. Earning this token requires you to have over 170 Max Life at the final encounter so use Encounters such as The Maiden, Man Eating Plant and The Alchemist as well as equipment such as the Imperial Armour and The Shield of Aegle.
Once you have the Reaper's Arrow simply replay the Death Challenge and ensure that you have the Reaper's Arrow equipped at the final encounter. This is easy to do using equipment heavy Encounters such as Thomas the Ogre and minimizing the number of Artifacts you are carrying.
The Reaper's Arrow will do damage of about 30% of the Reaper's total life per use. It comes with 3 uses total so you will need to do some damage yourself. This also worked for me on the assassin in The Hierophant (even though they're classed as a Silencer). I found it easier to get to, as you can skip all the clue stuff, shortcut to the end when you manage to pick up the Reapers Arrow. Then, just accuse anyone and you will end up fighting the real assassin anyway.

Also, in the Blacksmith quest line there is an encounter Terror-vein Ore, it also has an Assassin as an enemy.

I did this in the final challenge " The World". You can actually select the reapers arrow in your starting gear for this challenge. There is one assassin that you fight in the thieves wave.
I took out all the other enemies from this wave then about 75% of the assassains health. Then finished him with the reapers arrow.
It also may help to note that this can be obtained on easy mode, which makes the combats alot easier.

Easiest place to get this done is actually the Death encounter: the Reaper himself counts as an Assassin, and his final battle is him entirely alone. This works well because Reaper's Arrow can't be targeted, so you would otherwise need to kill everyone else and isolate and weaken an Assassin first.
