Kills that count toward this achievement include any kills you get while manning a turret, flying a jet, helicopter, or AC-130, or while operating a tank.
You should earn this achievement easily in your first playthrough of the game, and certainly by the end of the second playthrough. If for some reason you don't, go to mission 5 and use the AC-130 to kill some more enemies until you get it.
Note: sometimes the achievements in this game unlock immediately, sometimes they don't unlock until the end-of-mission screen, and sometimes they wouldn't unlock until I pressed the Xbox Guide button to go to the achievements tab.

These weapons consist of Turrets, Rocket Launchers, and other weapons that you are in control of throughout the game. You should have no problem unlocking this as you will use these in mostly every level of the game.
