The Achievement is called High Roller and you get it after you accumulate $5,000,000 on ONE of the difficulties.
You can easily get this in one play-through if you don't spend any money(think I got it with 2 or 3 levels to go). Try and go for Professional or Silent Assassin rating, you'll get extra $100,000-$150,000 per level.

I was spending during my playthrough so can I replay a mission several times to get the money?

This is not a solution, but a list of the maximum amount of money per mission you can earn without any glitches, if you achieved Silent Assassin Rating.
The following is based on Professional difficulty:
Starting Fund : $75,000
Mission 01 - Death of a Showman ----------- $300,000 (Professional Rating)*
Mission 02 - A Vintage Year ------------------- $550,000 (Silent Assassin Rating)
Mission 03 - Curtains Down ------------------- $550,000 (Silent Assassin Rating)
Mission 04 - Flatline ---------------------------- $520,000 (Silent Assassin Rating)
Mission 05 - A New Life ------------------------ $420,000 (Silent Assassin Rating)
Mission 06 - The Murder of Crows ----------- $620,000 (Silent Assassin Rating)

Payout might not vary between difficulties.
I know I finished A New Life on Rookie with a Silent Assassin rating and got $420,000, same as you on Professional. And from looking at how much money I have - 2.5m after A Murder of Crows, with only a few upgrade purchases - I think it might be the same for all other missions.

I know for sure I saved A New Life at the end of the level and kept reloading that save and eventually hit the 5 mli mark. Not sure how but it worked. I'm playing the new HD version on on X1 now and I'm trying to figure out how I did it.

If you want to get money for any of the weapon upgrades the fastest way is to do "A New Life" and get "Hitman" rating. Since you only get rating bonus once you've done the level, only the rating matters. Silent Assassin nets $150,000 but takes a lot longer than 2 minutes so the best thing to do is the method shown on the video below. I don't have a capture card but I do have the PC version so it's on PC.
