Walkthrough of Hitman Episode 6: Hokkaido on Silent Assassin.
This can be completed immediately with NO mastery required.
All targets are eliminated for the Silent Assassin challenge worth 5000xp - this will help towards mastery level 20. There are a number of other challenges completed that don't show because I'm already at level 20. If you're starting from scratch you'll get in the region of 10,000xp straight off (perhaps more).

Using this video on a first play through it gave me Mastery level 7 with the following challenges completed:

Story-related, can not be missed. You will unlock this upon completing the mission for the first time regardless of how you do it. I would suggest using two opportunities to kill the targets (Yuki Yamasaki + Erich Soders), since you will have to complete them all anyway.
