How to unlock the 1st Kill achievement in Hitman HD Pack - Definitive Guide

I'm going to post this solution for the following achievements: "Humanist", "1st kill", "Killed the wrong guy", and "Headshot". These can all be obtained within about 1 minute from the start of the very first real mission called "Anathema". Here's how:
Start up the mission and look down and to the left to see a mailman walking through a big driveway opening. He's your victim. I like to call him "John" He will walk up to deliver some flowers to the main gate but first John has to stop and see a man about a horse (he has to take a piss). This is where you strike.
Start out by equipping your anesthetic right from the start. Now start jogging towards John and when you get close enough (say about 10ft or so from him) hit Lb and enter sneak mode so he doesn't hear your approach. Keep following but wait for him to stop and pee otherwise you will never catch him as he walks faster than you can in sneak mode and without entering sneak mode you get busted everytime. Don't worry about how slow 47 is because John pees for quite awhile. Anyway as John's draining the snake creep close enough and hit him with the anaesthetic (be sure to hold Rt so he gets the fully sedated) there's your first achievement "humanist" secret achievement for successfully using anaesthetic. Now pull out your ballers and line him up for a nice unconscious head shot. When your ready for John to never see his family again (he has a family) plug him in the head and you should get 3 more achievements- "1st kill", "Killed the wrong guy" (for killing an innocent) and lastly "headshot".
There you go 4 very easy achievements within a minute of starting the first mission. Now if that's not how you wanted to play it out just restart the mission...no harm no foul.

Tried your way a few times with no luck, John kept running away and I was sneaking and everything, so I got fed up with restarting the mission and just bum rushed him at the gates first set of trees and was able to take him down.. before he started watering the trees..

This achievement simply entails that you kill someone. It doesn't matter if it is a guard, a civilian or a target. Just kill them in anyway. If you want this out of the way immediately, then simply kill a fellow inmate or a SWAT member on the first level with any method.
If you want to unlock this with 2 other achievements, take a pistol from the start and get a head-shot on one of your fellow patients. This will unlock you an achievement for getting a kill, killing an innocent AND getting a head-shot. 3 achievements in less than 3 seconds.
