I'm going to post this solution for the following achievements: "Humanist", "1st kill", "Killed the wrong guy", and "Headshot". These can all be obtained within about 1 minute from the start of the very first real mission called "Anathema". Here's how:
Start up the mission and look down and to the left to see a mailman walking through a big driveway opening. He's your victim. I like to call him "John" He will walk up to deliver some flowers to the main gate but first John has to stop and see a man about a horse (he has to take a piss). This is where you strike.
Start out by equipping your anesthetic right from the start. Now start jogging towards John and when you get close enough (say about 10ft or so from him) hit Lb and enter sneak mode so he doesn't hear your approach. Keep following but wait for him to stop and pee otherwise you will never catch him as he walks faster than you can in sneak mode and without entering sneak mode you get busted everytime. Don't worry about how slow 47 is because John pees for quite awhile. Anyway as John's draining the snake creep close enough and hit him with the anaesthetic (be sure to hold Rt so he gets the fully sedated) there's your first achievement "humanist" secret achievement for successfully using anaesthetic. Now pull out your ballers and line him up for a nice unconscious head shot. When your ready for John to never see his family again (he has a family) plug him in the head and you should get 3 more achievements- "1st kill", "Killed the wrong guy" (for killing an innocent) and lastly "headshot".
There you go 4 very easy achievements within a minute of starting the first mission. Now if that's not how you wanted to play it out just restart the mission...no harm no foul.

This is a good solution, but the headshot achievement doesn't seem to pop :(. +1 though

Possibly due to the games glitchy shooting maybe? I know 47 kinda shoots funny in silent assassin. Often I would aim accurately at someones head and end up shooting over or to the side of my target. If u aimed gor the head and accidently your bullet hit the chest kinda thing. Or the achievement is buggy im not sure but this is exactly how I popped it. Not like its very hard to just headshot someone anyway in any level.

This achievement simply entails that you get a headshot on any person in the game. It can be your target, or a guard, or a civilian. If you want to get this out the way immediately, then just pick up a pistol on the first level (The asylum) and aim at one of the fellow patient's heads and pull the trigger.
If you press Right stick in, you enter a First-person mode, which can make aiming easier throughout the campaign.
