I found it easiest to do this Achievement on Mission 1 The Hoodlum Dolls Stage 1(Sweet Angel)
All you have to do is not pick up any weapons(which is done with the B Button)
Just don't use the B button the whole stage , don't use any weapons other than your starting weapon and once you kill the boss the Achievement will unlock.
There is no need for a boss strategy as he is so easy due to being the first boss in the game. Keep your distance and pick up health when running low.
