It's hard to give a very detailed guide since you can craft your deck how you want it, but it seems most of the boss battles have a trick to them to win. In this case, you want to kill the prospector's pack mule in the first round of the two round boss battle. Just be aware that it moves to an adjacent space after every turn - it doesn't stay put. I won't go into detail, but you'll gain a significant advantage when you kill the pack mule.

The Prospector is the first of four bosses you will come up against in Act I. If you have not lost your first candle flame yet for any boss fight, your opponent will take it out and give you a The Smoke card that has no cost, zero Attack, 1 HP and will award four bones when attacked or sacrificed.
The Prospector first places his Pack Mule card on the left side and a attacking card to the right of it (Usually a Coyote - 2 Attack, 1 HP) in the second row. Your first move should be to focus on taking out the Pack Mule's five hit points as well as defending slot 2. Slot 3 will get an attacking card in the next turn, Slot 4 should have a 5 HP Boulder helping protect you.
Once you take out the Pack Mule you will be given all the cards hiddern in the Mule's pack and will have more than enough good cards to end the first of two rounds.
In Round Two, The Prospector will hit any cards you still have in play with his pickaxe and turn it into a 2 HP Golden Nugget. You will have to wait until your opponent takes out these nuggets to play new cards (they can not be sacrificed), but the line of defensive cards gives you a chance to grab more Squirrels or more of your deck. Once the way is clear, clean house and knock the Prospector out with your best cards to end the battle with a win and this trophy.
