To start, this is MUCH easier in Endless mode. Doing it in the main game is possible, but I wouldn't recommend it.
100x8 is achieved when you pick up eight of the enemy food drops in a row. There is a short window of time to collect them and keep the multiplier going. Once you hit 100x8 for the first time, you'll keep the combo until you take too long to pick up an item drop. I got this around wave 7/8. Play through a bit of the main game first so you'll have some better weapon pick ups. When you get one of the better ones, just clear the screen of enemies the best you can and start collecting! You should be able to knock a good amount of them out in the first couple rounds, but by wave 7+ you should be able to get it with ease.

I was going for the beat 10 levels cheeve and got this one, might be level 8 - 12, depends how fast you pick the goodies up and how good you are at the game. Thumbs up from me.

The roadmap has a more detailed explanation of 100x8s, how they work, and how they're critical to success in this game. As the description says, you need to get 100 of these in a single run.
This will likely sound pretty intimidating. It certainly did to me. However, I got this achievement naturally on Floor 3 of my successful New Game run, meaning I still have a whole other floor still to go. You should be able to make some progress through the first two floors, but where your 100x8 count really increases by a huge amount is on Floor 3 when you encounter the worm enemies. These enemies require you to shoot them enough to break off all body pieces one by one until they finally die, but because each body piece functions like an enemy, you can usually get 3-4 food pieces dropped per worm, making it very easy to build up to 100x8 and then maintain it in these rooms. You can easily get 10-20 100x8s in a single one of these worm rooms on Floor 3, so make sure you take full advantage of them and you'll get this achievement no problem.
