This is the only achievement affected by difficulty. What you need to do is set the game to Hard difficulty on the main menu and then load up either New Game or NG+, and then just beat the first floor.
I recommend waiting to do this until after you've at least beaten New Game mode, but preferrably after NG+ as well. You'll have enough experience by then that this won't be much of a challenge. You'll also have unlocked tons of new weapons that'll make this easier. The first few rooms won't even have a noticeable difference, and enemies will only be a bit tougher to kill in later rooms. You'll notice the biggest difference with how long it takes to kill the boss, but even then, all attack patterns are the same and your health is the same. So, just use all the same strategies you used on Easy for beating runs and you should have no problem clearing just the first floor on Hard.
