Endless Mode unlocks after finishing your first run (win or lose), and is accessed from the main menu. It's just you in a single room, where wave after wave of enemies come after you. They are increasingly more difficult, similar to progressing through the rooms and floors of the regular game. The main different here is that, since you're in a single room, there are no shops to buy stat upgrades. Your only source of stat upgrades is therefore building and maintaining 100x8 to get a stat upgrade to drop.
Don't let that thought stress you out though, as in my experience, getting through 10 waves felt like the equivalent of getting through two floors in the regular game, in terms of enemy difficulty. So, if you've already beaten the game before attempting this, ten waves will be no problem, especially with all of the weapons and such you've unlocked. Remember this can be done on Easy as well.
