To clear the mission Island Rampage you must defeat 100 enemies. The majority of the enemies only take one hit to kill until the last five or so.
The batch of normal enemies are easy, they aren't very offensive and only take one hit, however the last ones you need to be careful of as it's infuriating to get that far and die.
The way I did this was I used Okita Soji and used his 3-hit combo: B-B-B and before the third hit landed I'd block to cancel it and start the combo again. You need to watch out when it's the last few enemies though. Also I play the game very offensively whereas you might not so this might not be good for you.
Also you only take one hit to be killed.
