Must be done in a career game mode.
Easiest time to do so is at the very beginning of the career mode. Once the Space Centers loads in find the building called 'Mission Control'. Select either the "Collect scientific data from Kerbin" or "Launch your first vessel" contracts (or both as you're allowed 2 active contracts at a time until you upgrade the Mission Control building).
Collecting science can be done by pressing
on a science part while in control of the respective craft and selecting the 'log/observe' option and, either keeping the report and recovering the craft, or, transmitting the data right away (assuming the craft has some sort of antenna)(this option also yeilds less science). Another option is to press on a command module, selecting the 'crew report' option and either keeping or transmitting the data.
Launching a vessel is the name of the game and can be done by simply pressing
(with cursor mode disabled (clicking in on the 
swaps between the 2 modes)) while on the launchpad with any craft. Doing this will also grant you
