To perform an Ultra, Your target has to be in his blink red bar and you must start a combo before the ultra will work.

THIS IS HOW I DID IT (DOES NOT WORK IN TRAINING - need 2 controllers):
Go to Local Play, Select TUSK.
On 2nd controller, press A, select anyone.
use Tusk to kill opponent. One 2nd healthbar, get them down to the RED blinking health.
Start a combo:

Then immediately start Tusk's ULTRA COMBO (use the Left Thumbstick, not DPAD):

- according to other comments "A" is not correct.
+ - if "A" doesn't work, try Y. My mistake.
I got it my first try after wasting TONS of time with other characters, other moves, other combos...
