Hisako can appear in free rotation, you may have to wait for her... Load up a survival match and have at it, remember to adjust your difficulty or set up accordingly. Play through it in one sitting and in no time this should unlock. The objective is to beat 10 opponents first, it may take anywhere from 10-20 rounds to actually win. Just go as far as you can and it should unlock in no time.
