It is better to do this trophy almost last, because it is advisable to complete all the side quests, they often give Spheres.

There are two types of monsters and there are many of them nearby, each often dropping a Soul. The tactics are simple, go to the Southern Swamp, cast something massive (Scarlet Meteor is ideal), collect loot and go to the Western Swamp, clear the first island, return back to the Southern Swamp.
A little less than an hour and 180 Souls fell.
